Mouse chemokine ligand 1 (CCL1, TCA-3) is a 9 kDa secreted CC-chemokine whose N-terminal loop and 3₁₀ helix engage CCR8 on activated T-cells and macrophages, steering Th2 recruitment. A solvent-exposed 12-aa "SKP" surface (aa 30-41) between β2 and β3 strands folds into a β-hairpin unique among murine CCL proteins, presenting a conformational epitope preserved after glycosylation and fixation. Affinity-purified antibodies targeting this loop trace native CCL1 in thymic medulla, lung eosinophil foci, and mast-cell granules, delivering crisp extracellular deposition and single-band clarity on immunoblots with no cross-reaction to CCL2 or CCL3.
