Target Information
Mouse caspase 12 is a 373-residue inflammatory caspase comprising an N-terminal CARD (caspase recruitment domain), a central catalytic subunit with the conserved QACRG active-site pentad, and a C-terminal region that undergoes proteolytic removal during activation. The protein localizes to the endoplasmic reticulum outer membrane via its CARD, where it forms filaments through homotypic CARD interactions. Constitutively expressed in liver, kidney and lung, caspase 12 cycles between inactive monomers and activated oligomers. Antibodies raised against the CARD or large-subunit peptide detect native, cleaved and polymerized forms with negligible cross-reactivity to caspase 1 or 11, enabling selective immunoblot and immunofluorescence visualization in mouse tissue homogenates and cultured cells.
