Mouse calcium/calmodulin-dependent protein kinase II alpha (CaMKIIα) is a 54 kDa serine/threonine kinase whose N-terminal kinase domain, autoinhibitory segment, and variable linker assemble into a 12-subunit holoenzyme that decodes Ca²⁺ oscillations. A solvent-exposed 16-aa "KVE" loop (aa 280-295) between the regulatory and association domains folds into a β-hairpin unique among mouse CaMKII isoforms, presenting a conformational epitope preserved after autophosphorylation and fixation. Affinity-purified antibodies targeting this loop trace native CaMKIIα in hippocampal synapses, cortical dendrites, and cardiac sarcoplasmic reticulum, delivering crisp punctate staining and single-band clarity on immunoblots with no cross-reaction to CaMKIIβ or CaMKIIδ.
