Target Information
Mouse breast cancer susceptibility protein 1 (BRCA1) is a 1,812-residue nuclear phosphoprotein harboring an N-terminal RING finger with E3 ubiquitin ligase activity, two centrally located BRCA1 C-terminal (BRCT) repeats that bind phosphorylated peptides, and multiple SQ/TQ clusters targeted by ATM/ATR kinases. The polypeptide assembles into heterodimers with BARD1, stabilizing the RING domain and enhancing ligase catalysis. Expression peaks during S and G2 phases, localizing to DNA damage foci, centrosomes and replication forks. Antibodies generated against full-length or peptide epitopes of mouse BRCA1 detect both native and phosphorylated isoforms with negligible cross-reactivity to BRCA2 or other RING proteins, enabling specific visualization in nuclear extracts and fixed tissue sections.
