Mouse ATPase, Na+/K+ transporting alpha 1 polypeptide (ATP1A1) is a 113 kDa type-II membrane protein whose ten transmembrane helices form the catalytic core that exchanges three Na+ for two K+ against electrochemical gradients using ATP hydrolysis. A solvent-exposed 14-aa "KVE" loop (aa 710-723) between transmembrane segments 7 and 8 folds into a β-hairpin unique among mouse P-type ATPases, presenting a conformational epitope preserved after membrane isolation and fixation. Affinity-purified antibodies targeting this loop trace native ATP1A1 in basolateral membranes of nephron tubules, choroid plexus, and cardiac sarcolemma, delivering crisp lateral rings and single-band clarity on immunoblots with no cross-reaction to alpha 2 or alpha 3 isoforms.
