Target Information

Mouse anoctamin 1 (ANO1) is an 887-residue eight-span membrane protein that assembles as a dimer to form Ca²⁺-activated chloride channels. Each subunit contains a conserved "E-EEEE" cytoplasmic motif that coordinates cytosolic Ca²⁺ and triggers pore opening via helix repositioning within transmembrane segments 6 and 7. Constitutively expressed in airway epithelium, salivary glands, dorsal root ganglia and interstitial cells of Cajal, ANO1 localizes to plasma membrane and intracellular vesicles. Antibodies raised against the third extracellular loop or C-terminal tail detect native glycosylated and unglycosylated isoforms with negligible cross-reactivity to ANO2-ANO10, enabling selective immunoblot, immunohistochemistry and flow-cytometric analysis in mouse tissue homogenates and cultured cells.
