Target Information
Mouse angiotensin I converting enzyme (ACE, CD143) is a 130 kDa type-I dipeptidyl carboxypeptidase whose two homologous catalytic domains cleave angiotensin I and bradykinin, regulating blood pressure and inflammation. A solvent-exposed 14-aa "KVE" loop (aa 620-633) within the C-domain active site folds into a β-hairpin unique among mouse metallopeptidases, presenting a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native ACE in pulmonary capillary endothelium, renal proximal tubules, and splenic follicles, delivering crisp apical membranes and single-band clarity on immunoblots with no cross-reaction to ACE2 or other peptidases.
