Mouse angiopoietin-like protein 8 (ANGPTL8) is a 22 kDa secreted peptide whose N-terminal coiled-coil clamps onto ANGPTL3 to unleash its lipoprotein lipase-inhibitory arm, while a C-terminal fibrinogen-like stub remains disordered until complex formation. A unique 14-residue "QRK" insert (aa 50-63) interrupts the first helix, creating a conformational epitope absent from other ANGPTL members. Polyclonal antibodies targeting this insert trace native ANGPTL8 in liver sections, adipose tissue, and serum, yielding crisp cytoplasmic granules and single-band clarity on immunoblots with no cross-reaction to ANGPTL3 or ANGPTL4.
