Target Information
Mouse alpha-1-antichymotrypsin (Serpina3) is a 398-residue, 68 kDa secreted glycoprotein belonging to the clade A serpin superfamily. Synthesized primarily in hepatocytes and activated astrocytes, the molecule presents a mobile reactive-center loop (P1-P1' = Leu-Ser) that traps target chymotrypsin-like proteases through rapid formation of a 1:1 stable acyl-enzyme complex followed by loop insertion into β-sheet A. Two N-linked glycans shield the hydrophobic breach region, while a single disulfide bridges residues 256-277. Antibodies raised against native or peptide epitopes of mouse Serpina3 detect intact, cleaved and polymerized forms with negligible cross-reactivity to Serpina1 or other serpins, enabling precise immunoblot and immunohistochemical localization in mouse plasma and tissue sections.
