Mouse adenylate cyclase 6 (ADCY6) is a 116 kDa 12-span membrane enzyme whose N-terminal VVLQ motif and two cytoplasmic catalytic loops convert ATP to cAMP under Gαi/q control. A solvent-exposed 18-residue "EIK" helix (aa 495-512) in the C1b domain projects between membrane bilayers, presenting a conformational epitope unique among ADCY isoforms. Affinity-purified antibodies targeting this helix trace native ADCY6 in cardiomyocyte sarcolemma, airway smooth muscle, and neuronal dendrites, delivering crisp membrane rings and single-band clarity on immunoblots with no cross-reaction to ADCY5 or ADCY8.
