Guinea-pig prealbumin (transthyretin) is a 55 kDa homotetramer whose subunits sandwich two thyroxine sites between paired β-barrels. A surface-exposed DE-loop (aa 102-115) carries a unique Cys-Pro motif absent from rodent or human orthologs, creating a strain-specific epitope that resists tetramer dissociation. Antibodies raised against this loop detect native prealbumin in serum, cerebrospinal fluid, and choroid plexus, yielding crisp single-band clarity on immunoblots and crisp apical staining with no cross-reaction to albumin or retinol-binding protein.
