Target Information

Chicken complement component 4 (C4) is a 1,730-residue plasma glycoprotein synthesized as a single-chain proprotein that undergoes proteolytic maturation into three disulfide-linked chains: α (93 kDa), β (70 kDa) and γ (30 kDa). The α-chain contains a buried thioester bond (Cys-Gly-Glu-Gln) that becomes exposed upon C1s cleavage, allowing covalent attachment to target surfaces. Constitutively expressed in liver and secreted into serum, C4 localizes to immune complexes and cell membranes during activation. Antibodies raised against the α-chain or thioester-containing region detect native, cleaved and activated forms with negligible cross-reactivity to C3 or C5, enabling selective immunoblot and immunohistochemical visualization in chicken plasma and tissue sections.
