Target Information
Bovine cathepsin S is a 24 kDa lysosomal cysteine protease whose active-site cleft accommodates bulky hydrophobic side chains, cleaving invariant chain and extracellular matrix proteins at neutral pH. A solvent-exposed "GNE" loop (aa 152-160) within the L-domain exhibits a bovine-specific glycine insertion, presenting a conformational epitope preserved in both pro- and mature forms. Affinity-purified antibodies targeting this loop detect native cathepsin S in alveolar macrophages, lymph node follicles, and milk exosomes, yielding crisp vesicular staining and single-band clarity on immunoblots with no cross-reaction to cathepsin B or cathepsin L.
