Target Information

The non-selective voltage-gated ion channel VDAC1 facilitates the transport of anions and cations across both the mitochondrial outer membrane and the plasma membrane. In the outer mitochondrial membrane, it permits the diffusion of small hydrophilic molecules, while in the plasma membrane, it plays a role in cell volume regulation and apoptosis. VDAC1 adopts an open conformation at low or zero membrane potential and a closed conformation at potentials above 30-40 mV. The open state exhibits weak anion selectivity, whereas the closed state is cation-selective. In skeletal muscle, the absence of VDAC1 reduces mitochondrial sensitivity to ADP, impacting the muscle's metabolic capacity. Research indicates that mice lacking VDAC1 experience muscle fatigue and mitochondrial dysfunction, highlighting VDAC1's crucial role in maintaining muscle energy metabolism and motor function.
