Nicotinamide phosphoribosyltransferase facilitates the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to produce nicotinamide mononucleotide, an intermediate in NAD biosynthesis. Its secreted form acts as a cytokine with immunomodulating properties and an adipokine with anti-diabetic effects, though it lacks enzymatic activity due to the absence of ATP activation, which is scarce in extracellular space and plasma. In mice, Nampt has diverse functions, including roles in energy metabolism, cell survival, inflammatory response, neuroprotection, and tumor immunity. In mouse adipose tissue, the absence of Nampt significantly impairs the heat production capacity of brown and white adipose tissues, leading to reduced adaptive responses to cold exposure and β-adrenaline stimulation.
