Vascular endothelial growth factor receptor 3 (VEGFR-3) is a tyrosine-protein kinase that serves as a cell-surface receptor for VEGFC and VEGFD, playing a crucial role in adult lymphangiogenesis and the development of the vascular network and cardiovascular system during embryonic development. It facilitates the proliferation, survival, and migration of endothelial cells, while also regulating angiogenic sprouting. VEGFR-3 is a key regulator of lymphangiogenesis, with its ligands VEGF-C and VEGF-D promoting the formation and maintenance of lymphatic vessels through VEGFR-3 activation. For instance, in mice, the absence of VEGFR-3 during embryonic development results in abnormal vascular tissue and cardiac dysfunction, whereas in adulthood, VEGFR-3 is primarily expressed in lymphatic endothelial cells. Additionally, the activation of VEGFR-3 involves downstream signaling pathways like ERK and AKT, which are vital for the proliferation, migration, and survival of lymphatic endothelial cells.
