Target Information

Tyrosine Hydroxylase catalyzes the transformation of L-tyrosine into L-dihydroxyphenylalanine (L-Dopa), serving as the rate-limiting step in the synthesis of catecholamines, including dopamine, noradrenaline, and adrenaline. This process utilizes tetrahydrobiopterin and molecular oxygen. Tyrosine 3-hydroxylase (TH) is crucial in the biosynthesis of catecholamines such as dopamine, norepinephrine, and epinephrine, highlighting its significant biological role in mice. TH facilitates the conversion of tyrosine to 3,4-dihydroxyphenylalanine (DOPA), marking the initial and rate-limiting step in catecholamine synthesis, thus playing a vital role in regulating these neurotransmitters. In mice, mutations in the TH gene can result in severe physiological impacts. For instance, knockout of the TH gene leads to a significant reduction in TH activity in both embryonic and adult tissues, though catecholamine levels are maintained in adulthood through compensatory regulatory mechanisms.
