Target Information
Tumor susceptibility gene 101 protein (TSG101) is part of the ESCRT-I complex, a crucial regulator of vesicular trafficking processes. It binds to ubiquitinated cargo proteins and is necessary for sorting endocytic ubiquitinated cargos into multivesicular bodies (MVBs). TSG101 facilitates the interaction between the ESCRT-0 and ESCRT-I complexes. It is vital for cell growth, proliferation, and survival in both embryonic and adult tissues. In mouse embryos, the absence of TSG101 results in developmental arrest and failure to complete implantation, leading to early embryonic death. Additionally, TSG101 plays an important role in adult tissues; for instance, in mammary epithelial cells, its loss causes cell cycle arrest and p53-dependent cell death. TSG101 is potentially involved in various diseases. In neurodegenerative conditions, abnormal regulation of TSG101 is linked to the development of spongiform encephalopathy in mice, possibly due to defects in endosomal trafficking functions.
