The Tumor Necrosis Factor Receptor Superfamily Member 4 (also known as TNFRSF4 or OX40) acts as a receptor for TNFSF4/OX40L/GP34. This costimulatory molecule plays a crucial role in sustaining long-term T-cell immunity. In mice, TNFRSF4 is primarily involved in regulating immune responses and facilitating the activation and differentiation of T cells. It is particularly significant in the activation of CD4+ T cells, contributing to the development of both effector and memory T cells. Research indicates that the TNFRSF4 signaling pathway enhances T cell proliferation and differentiation while boosting their antigen responsiveness. For instance, activating TNFRSF4 can augment antigen-specific clonal expansion of CD4+ T cells post-vaccination, thereby strengthening the immune response.
