Target Information
Tumor necrosis factor receptor superfamily member 1A acts as a receptor for TNFSF2/TNF-alpha and homotrimeric TNFSF1/lymphotoxin-alpha. The adapter molecule FADD recruits caspase-8 to the activated receptor, forming a death-inducing signaling complex (DISC) that activates caspase-8 and triggers a cascade of caspases (aspartate-specific cysteine proteases), thus mediating apoptosis. In mice, TNFR1, encoded by the Tnfrsf1a gene, plays a critical role in regulating immune responses, inflammation, and apoptosis through participation in TNF signaling. Mouse models show that the expression level of TNFR1 significantly influences TNF-mediated inflammatory responses and cell death. In intestinal epithelial cells, the level of TNFR1 expression is crucial in determining the extent of TNF-induced cell death.
