Tumor necrosis factor receptor superfamily member 17 serves as a receptor for TNFSF13B/BLyS/BAFF and TNFSF13/APRIL, promoting B-cell survival and aiding in the regulation of humoral immunity. It activates NF-kappa-B and JNK. In mice, TNFRSF17 has several critical functions, including maintaining the survival of plasma cells, regulating antibody responses, participating in immune regulation and disease processes, and influencing the tumor microenvironment and intestinal immunity. TNFRSF17 is crucial for bone marrow plasma cell (PC) survival, as studies reveal its necessity for plasma cell longevity via transmembrane activation and cyclosporine L binding receptor-mediated signaling pathways. In TNFRSF17 knockout mice, plasma cell survival is significantly reduced, demonstrating the receptor's vital role in sustaining long-term plasma cell survival.
