Target Information
Tumor necrosis factor receptor superfamily member 11A serves as a receptor for TNFSF11/RANKL/TRANCE/OPGL and is crucial for RANKL-mediated osteoclastogenesis. In mice, knocking out the Tnfrsf11a gene results in severe osteopetrosis due to the inability to form osteoclasts. Additionally, RANKL (TNFSF11) facilitates osteoclast differentiation and activity by activating the RANK (TNFRSF11A) signaling cascade, thus supporting bone remodeling. On one hand, excessive RANKL expression may foster tumor growth and metastasis. In mouse models, deletion of Tnfrsf11a led to reduced tumor growth and inhibited liver metastasis of HeLa cells. On the other hand, excessive activity of TNFRSF11A might be linked to certain cancers. KLF5 enhances the proliferation, migration, and invasion of cervical cancer cells by modulating TNFRSF11A expression.
