Target Information
Tumor necrosis factor ligand superfamily member 4 is a cytokine that interacts with TNFRSF4 to co-stimulate T-cell proliferation and cytokine production. Tnfsf4 is predominantly expressed by B cells, particularly in synovial macrophages (SMs). In the rheumatoid arthritis (CIA) model, the absence of Tnfsf4 results in abnormal differentiation of T follicular helper (Tfh) cells. Similarly, in the systemic lupus erythematosus (SLE) model, the loss of Tnfsf4 leads to diminished T cell activation, higher proportions of central memory and naive T cells, and reduced proportions of CD4+ Tfh cells. In a bacterial-induced bone resorption model, Tnfsf4 expression showed sex-specific differences in female and male mice. For instance, in CD11bhi cells, the expression of NFATc1 was notably higher in female mice than in male mice on day 2, whereas Tnfsf4 expression was greater in male mice.
