Tumor Necrosis Factor Ligand Superfamily Member 14 (TNFSF14) is a cytokine that binds to TNFRSF3/LTBR and modulates its effects through the decoy receptor TNFRSF6B. It activates NFKB and stimulates T-cell proliferation. As a ligand for TNFRSF14/HVEM, it delivers costimulatory signals to T cells upon binding, leading to T-cell proliferation and IFNG production. TNFSF14 plays a significant immunomodulatory role by interacting with its receptors, HVEM (herpes virus entry mediator) and LTβR (lymphotoxin β receptor). Research indicates that TNFSF14 is crucial in intestinal inflammation in mice; for instance, in the DSS-induced acute colitis model, mice lacking TNFSF14 exhibited significantly reduced inflammatory responses.
