Target Information
Tumor necrosis factor ligand superfamily member 12 interacts with FN14 and potentially with TNRFSF12/APO3. It serves as a weak inducer of apoptosis in certain cell types and mediates the activation of NF-kappa-B. This protein encourages angiogenesis and the proliferation of endothelial cells and is involved in the induction of inflammatory cytokines, promoting IL8 secretion. In mice, Tnfsf12 (TWEAK) functions predominantly as tumor necrosis factor superfamily member 12, with roles connected to various pathological processes, especially atherosclerosis, vascular remodeling, and muscle pathology. Mice lacking the Tnfsf12 gene exhibited significantly reduced atherosclerotic lesion areas and decreased lipid content in plaques in the diabetic ApoE knockout mouse model. Additionally, Tnfsf12-deficient mice showed milder pathological changes following vascular injury, including reduced intima/media areas and lower cell density.
