Target Information
The tumor necrosis factor-inducible gene 6 protein plays a crucial role in regulating the organization of the extracellular matrix during the process of tissue remodeling. It facilitates the transfer of a heavy chain (HC) from the inter-alpha-inhibitor (I-alpha-I) complex to hyaluronan. This involves cleaving the ester bond between the C-terminus of the HC and the GalNAc residue of the chondroitin sulfate chain within the I-alpha-I complex, followed by the transesterification of the HC to hyaluronan. This mechanism enhances the antiprotease function of the I-alpha-I complex by releasing free bikunin. Mice that lack TNFAIP6 show intensified inflammatory responses in antigen-induced arthritis, characterized by more pronounced synovial infiltration, destruction of articular cartilage, and bone erosion.
