Target Information
In mice, tryptophan hydroxylase 2 (Tph2) serves as the rate-limiting enzyme for synthesizing 5-hydroxytryptamine (5-HT) in the central nervous system. Tph2 is primarily expressed in the raphe nuclei of the brainstem and is responsible for catalyzing the conversion of tryptophan to 5-hydroxytryptophan, a crucial step in 5-HT synthesis. Functional defects in Tph2 result in substantially decreased 5-HT levels in the brain, which are closely associated with various behavioral and mood disorders. For instance, Tph2 knockout mice display symptoms such as depressive-like behavior, anxiety-like behavior, and increased aggression. Additionally, mice with diminished Tph2 function also exhibit resistance to specific drug treatments, including antidepressants.
