Target Information

Transcription Factor A, Mitochondrial (TFAM) interacts with the mitochondrial light strand promoter, playing a crucial role in regulating mitochondrial transcription. It is essential for mtDNA replication and transcription, and its deficiency results in profound mitochondrial dysfunction. For instance, mice with a complete TFAM knockout at the whole-body level experience embryonic lethality between E8.5 and E10.5 due to the absence of TFAM protein and a marked reduction or complete loss of mtDNA. Mice with a TFAM knockout specifically in skeletal muscle show significant mitochondrial impairment, characterized by a decreased number of mitochondria, lower mitochondrial DNA copy number, and disrupted function of the respiratory chain complexes.
