Target Information
TNF receptor-associated factor 6 (TRAF6) is an E3 ubiquitin ligase that, in partnership with UBE2N and UBE2V1, facilitates the formation of 'Lys-63'-linked polyubiquitin chains attached to proteins such as ECSIT, IKBKG, IRAK1, AKT1, and AKT2. TRAF6 is crucial for the activation, differentiation, and maintenance of T cells. A deficiency of TRAF6 in T cells can result in flawed Treg cells, potentially leading to autoimmune diseases. For instance, mice with TRAF6ΔT cells exhibit resistance to Treg suppression, causing excessive activation of Tconv cells and resulting in autoimmune inflammation. In B cells, TRAF6 is involved in CD40-mediated signaling and influences B cell activation, proliferation, and differentiation. While its absence minimally impacts the development of early B cells, it has a significant effect on the pool of mature circulating B cells.
