Thymosin beta-4 is crucial for cytoskeleton organization, as it binds to and sequesters actin monomers (G actin), thereby inhibiting actin polymerization. Thymosin β4 (Tβ4) is a 4.9 kDa actin-binding peptide, consisting of 43 amino acids, and is widely distributed across various tissues and cells, such as the skin, brain, lungs, liver, and myocardium. Tβ4 is pivotal in the development of mouse hair follicles. Research indicates that Tβ4 enhances hair growth by activating hair follicle stem cells and their progeny, facilitating their migration to the hair follicle base, and promoting follicle differentiation and remodeling. For instance, mice overexpressing Tβ4 exhibited accelerated hair growth and denser hair follicles. In the mouse skin, Tβ4 overexpression significantly boosted the migration and angiogenesis of vascular endothelial cells, which was linked to increased VEGF expression.
