Target Information

Talin-1 is a large cytoskeletal protein that is concentrated at sites of cell-matrix and cell-cell interactions. It plays a critical role in linking major cytoskeletal structures to the plasma membrane. As a key regulator of integrin activation, Talin-1 facilitates integrin engagement and binding to the extracellular matrix by interacting with the tail of the integrin β chain. In mice, a deficiency in Talin-1 impairs platelet integrin activation, leading to defective aggregation and thrombus formation. This results in severe bleeding complications but offers resistance to atherosclerosis. Additionally, in mouse models, the expression level of Talin-1 is closely associated with inflammation and tumor development. For instance, the absence of Talin-1 reduces the recruitment of mucosal macrophages, thereby providing protection against colitis.
