Sphingosine 1-phosphate receptor 1 (S1PR1) is a G-protein coupled receptor for the bioactive lysosphingolipid sphingosine 1-phosphate (S1P), typically associated with the G(i) subclass of heteromeric G proteins. This signaling pathway activates RAC1, SRC, PTK2/FAK1, and MAP kinases. S1PR1 plays a vital role in cell migration, likely by reorganizing the actin cytoskeleton and forming lamellipodia in response to stimuli that enhance sphingosine kinase SPHK1 activity. In mice, S1PR1 is involved in various functions such as angiogenesis, immune regulation, tumor progression, neuroprotection, and cardiac function. In mouse models, the absence of S1PR1 results in abnormal expansion and increased branching of blood vessels, along with reduced migration and proliferation of endothelial cells, thus impacting embryonic development and adult vascular function. S1PR1 also helps maintain vascular barrier integrity and prevent vascular leakage by regulating endothelial cell signaling pathways, including Ras-ERK and PI3K-Akt.
