Target Information

The proton-coupled zinc antiporter SLC30A5, in combination with SLC30A6, forms a functional antiporter that facilitates zinc entry into the lumen of organelles along the secretory pathway. By maintaining zinc ion homeostasis within the early secretory pathway, it regulates the activation and folding of enzymes such as alkaline phosphatases and those involved in the biosynthesis of phosphatidylinositol glycan anchors. In mice, SLC30A5 influences various functions, including bone health, cardiac function, zinc homeostasis, tumorigenesis, immune response, and mitochondrial activity. SLC30A5 is crucial for bone health, as studies have shown that mice lacking SLC30A5 develop severe osteoporosis, underscoring its role in bone development and density maintenance. Furthermore, SLC30A5 knockout mice exhibit cardiac abnormalities such as cardiomyopathy, fetal edema, and perinatal death, highlighting its importance in cardiac function and development.
