The proton-coupled zinc antiporter SLC30A1 functions as a zinc ion:proton antiporter, potentially operating at the plasma membrane to mediate zinc efflux from cells against its electrochemical gradient, thus protecting cells from intracellular zinc accumulation and toxicity. Alternatively, it might inhibit transport of the L-type calcium channels regulatory subunit, CACNB2, to the plasma membrane through a mechanism yet to be determined. In mice, Slc30a1 is found in the basement membrane of intestinal epithelial cells, where it facilitates zinc absorption and transport, playing a crucial role in maintaining systemic zinc homeostasis, particularly in the intestine. Studies using various tissue-specific knockout mouse models have shown that the absence of Slc30a1 leads to disruptions in intestinal zinc absorption, affecting overall zinc homeostasis and having severe consequences for survival.
