Target Information

Tumor necrosis factor ligand superfamily member 11 (also known as RANKL) is a cytokine that binds to TNFRSF11B (OPG) and TNFRSF11A (RANK). It enhances the ability of dendritic cells to stimulate the proliferation of naive T-cells and may serve as an important regulator of interactions between T-cells and dendritic cells, playing a role in regulating T-cell-dependent immune responses. Additionally, it may play a significant role in increased bone resorption associated with humoral hypercalcemia of malignancy. The Tnfsf11 gene is crucial for bone formation. Research has shown that Tnfsf11-/- mice exhibit abnormal distribution of osteoblasts and osteoclasts, with osteoblasts absent from normal areas and osteoclasts remaining stationary in these regions. Beyond bone formation, Tnfsf11 is important in immune and inflammatory responses. For instance, in a mouse asthma model, Tnfsf11 expression was significantly elevated and closely correlated with changes in inflammation-related biomarkers.
