Signal transducer and activator of transcription 5A (STAT5A) performs dual roles in both signal transduction and transcription activation. It mediates cellular responses to the cytokine KITLG/SCF and various growth factors, possibly including those activated by FGFR1, FGFR2, FGFR3, and FGFR4. STAT5A binds to the GAS element, triggering PRL-induced transcription and regulating the expression of milk proteins during lactation. It is crucial for the differentiation of mammary epithelial cells during pregnancy and lactation. Research indicates that the absence of STAT5A results in impaired formation and branching of mammary lobules, leading to mammary gland dysplasia. Moreover, STAT5A-deficient mice are unable to lactate due to inadequate mammary gland development, underscoring the critical role of STAT5A in mammary gland development and lactation.
