Target Information

Sideroflexin 1 functions as an amino acid transporter that imports serine into mitochondria, serving as a crucial substrate for the mitochondrial branch of the one-carbon pathway. Within mitochondria, serine is converted into glycine and formate, which then move to the cytosol to aid in the generation of charged folates acting as one-carbon donors. Sideroflexin-1 (SFXN1) is a multi-transmembrane protein, part of a conserved protein family evolutionarily, and is mainly expressed in mammals. In mice, mutations in Sideroflexin-1 result in pathological characteristics such as axial skeletal abnormalities, bent tails, and mitochondrial iron accumulation in erythrocytes, indicating problems with iron metabolism and heme biosynthesis. Additionally, mice lacking Sideroflexin-1 exhibit hypochromic and microcytic anemia, highlighting its critical role in iron metabolism.
