Target Information
Sestrin-2 acts as an intracellular leucine sensor that negatively regulates the mTORC1 signaling pathway via the GATOR complex. In the absence of leucine, it binds to the GATOR2 subcomplex, thereby inhibiting mTORC1 signaling. In mice, Sestrin-2 serves a wide array of functions, including maintaining metabolic homeostasis, providing antioxidant defense, offering neuroprotection, regulating the immune system, and suppressing tumors. In a mouse model of fatty liver induced by a chronic high-fat diet, Sestrin-2 helps to slow the progression from simple steatosis to metabolic-associated steatohepatitis (MASH) by inhibiting RIPK3-mediated necroptosis.
