Target Information
Serotonin N-acetyltransferase regulates the production of melatonin in the pineal gland, aligning it with the night/day cycle. It facilitates the N-acetylation of serotonin into N-acetylserotonin, the second-to-last step in melatonin synthesis. As a crucial enzyme in this process, AANAT converts serotonin into N-acetyl serotonin (NAS), a melatonin precursor. In mice, AANAT expression and activity are closely governed by circadian rhythms and are found not only in the pineal gland but also in the uterus. Research indicates that AANAT and the melatonin receptor MT2 are vital for successful early pregnancy in the mouse uterus. AANAT knockout (KO) mice exhibit reduced activity of endometrial receptors, increased expression of estrogen receptor α (ERα), and decreased expression of progesterone receptor (PR), leading to impaired endometrial receptivity.
