Target Information
Serine palmitoyltransferase 2 is a key component of the serine palmitoyltransferase (SPT) enzyme complex, which initiates and regulates the rate-limiting step in sphingolipid biosynthesis. It achieves this by condensing L-serine and activated acyl-CoA, typically palmitoyl-CoA, to produce long-chain bases. The SPT complex comprises SPTLC1, SPTLC2 or SPTLC3, along with SPTSSA or SPTSSB. The catalytic core of this complex is formed by the heterodimer SPTLC1 and SPTLC2/SPTLC3. By overexpressing Sptlc2, intrahepatic lipid droplet accumulation can be reduced, and insulin sensitivity improved, through enhanced secretion of VLDL (very low-density lipoprotein). Furthermore, overexpressing Sptlc2 decreases glucose production in the liver and enhances insulin signaling efficiency, thereby ameliorating insulin resistance induced by a high-fat diet.
