Serine palmitoyltransferase 1 is a component of the serine palmitoyltransferase (SPT) multisubunit enzyme, which catalyzes the initial and rate-limiting step in sphingolipid biosynthesis by condensing L-serine with activated acyl-CoA, typically palmitoyl-CoA, to form long-chain bases. In mice, Serine palmitoyltransferase 1 (SPT1) is integral to sphingolipid biosynthesis and plays crucial roles in cell membrane composition, cholesterol absorption, and the maintenance of cell morphology and function. Mice deficient in SPT1 exhibit reduced cholesterol absorption capacity, potentially linked to decreased expression of NPC1L1 (Niemann-Pick C1-like 1) and ABCG5/ABCG8 genes, both involved in cholesterol absorption and metabolism. Additionally, the absence of SPT1 results in a significant decrease in sphingomyelin levels in mouse hepatocytes, further impacting cholesterol absorption and metabolism.
