Target Information
Sepiapterin reductase is responsible for catalyzing the final one or two reduction steps in the biosynthesis of tetrahydrobiopterin, forming 5,6,7,8-tetrahydrobiopterin. In mice, the primary role of sepiapterin reductase (SPR) is in the biosynthesis of tetrahydrobiopterin (BH4). In a mouse model of inflammatory joint disease, inhibiting SPR can significantly reduce inflammation-induced pain. Research has demonstrated that SPR inhibitors can decrease inflammatory responses and lower both the thermal pain threshold and mechanical pain sensitivity in mice. SPR knockout mice (Spr−/−) experience cardiovascular issues such as hypertension, heart rate fluctuations, and bradycardia. These symptoms are linked to reduced BH4 levels, as BH4 plays a crucial role in regulating nitric oxide (NO) production.
