Target Information
Nicotinate-nucleotide pyrophosphorylase [carboxylating] plays a role in the catabolism of quinolinic acid (QA). In QPRT knockout (KO) mice, the absence of QPRT activity hinders effective QA degradation, leading to its accumulation in the body. This build-up can induce neurotoxicity, as QA is an excitotoxin for the NMDA receptor, potentially causing neuronal damage. Research has shown that QA levels significantly increase in the striatum of QPRT KO mice, accompanied by neuronal degeneration. These mice exhibit decreased motor and cognitive functions, such as reduced activity in the merry-go-round test, diminished spontaneous alternation behavior in the Y-maze test, and prolonged escape latency in the Barnes maze test.
