Target Information
Pyruvate kinase PKM catalyzes the final rate-limiting step of glycolysis by facilitating the transfer of a phosphoryl group from phosphoenolpyruvate (PEP) to ADP, thereby generating ATP. The balance between its highly active tetrameric form and the nearly inactive dimeric form determines whether glucose carbons are directed towards biosynthetic processes or utilized for glycolytic ATP production. This transition between the two forms is essential for controlling glycolysis and plays a critical role in tumor cell proliferation and survival. In mice, pyruvate kinase M (PKM) primarily functions as a key regulator in glycolysis and influences metabolic reprogramming across various tissues and disease states. PKM1 is predominantly expressed in tissues with high energy demands, like the brain and muscles, while PKM2 is broadly expressed in other tissues, including the liver, red blood cells, and certain cancerous tissues.
