Pyruvate Dehydrogenase Kinase Isozyme 4 is an enzyme that plays a crucial role in regulating glucose and fatty acid metabolism by phosphorylating the pyruvate dehydrogenase subunits PDHA1 and PDHA2. This action inhibits pyruvate dehydrogenase activity, thereby controlling the flow of metabolites through the tricarboxylic acid cycle, reducing aerobic respiration, and preventing the conversion of pyruvate to acetyl-coenzyme A. PDK4 facilitates the energy source shift from glucose to fatty acids by phosphorylating and inhibiting the mitochondrial pyruvate dehydrogenase complex (PDC). In mice, PDK4 expression is influenced by various physiological conditions, such as starvation, fasting, and insulin signaling. During periods of hunger, PDK4 expression increases, thus inhibiting PDC activity to conserve glucose and maintain blood glucose levels.
