Target Information
In mice, the β subunit of the pyruvate dehydrogenase complex (PDC) plays a crucial role in converting pyruvate into acetyl-CoA and carbon dioxide (CO2), linking the glycolysis pathway to the tricarboxylic acid (TCA) cycle. This conversion is a vital step in cellular energy metabolism, supplying ATP and supporting various metabolic activities. In the mitochondria of mouse skeletal muscle, the E1 component of PDC (comprising α and β subunits) is significantly enriched, underscoring its importance in muscle energy metabolism. Additionally, research indicates that PDC activity is regulated by pyruvate dehydrogenase kinase (PDHK), which modulates pyruvate metabolism by inhibiting PDC activity. This regulatory mechanism is altered in certain pathological models, such as MCT and SuHx models, where increased PDCβ mRNA levels and decreased PDHK4 expression are observed, potentially contributing to metabolic disorders.
