Target Information

DNA-dependent protein kinase is a serine/threonine-protein kinase that functions as a molecular sensor for DNA damage. It plays a crucial role in DNA non-homologous end joining (NHEJ), a process essential for repairing double-strand breaks (DSBs) and facilitating V(D)J recombination. Prkdc serves as a pivotal component of the NHEJ pathway, necessary for the repair of DNA double-strand breaks. This function is vital for lymphocyte development, as NHEJ is integral to V(D)J recombination, which underlies the diversity of T-cell and B-cell receptor genes. Additionally, Prkdc plays a significant role in maintaining mitochondrial DNA (mtDNA) in mice. Research has shown that mutations in Prkdc are linked to increased susceptibility to doxorubicin (ADR)-induced nephropathy in mice, resulting from the accumulation of mitochondrial DNA damage and mutations.
