Target Information
Protein kinase C alpha type (PKCα) is a serine/threonine-protein kinase activated by calcium, phospholipids, and diacylglycerol (DAG). It plays a role in both positively and negatively regulating various cellular processes such as cell proliferation, apoptosis, differentiation, migration, adhesion, cardiac hypertrophy, angiogenesis, platelet function, and inflammation. It does so by directly phosphorylating targets like RAF1, BCL2, CSPG4, TNNT2/CTNT, or by activating signaling pathways involving MAPK1/3 (ERK1/2) and RAP1GAP. In mice, PKCα has diverse functions including roles in visual processing, immune regulation, tumor suppression, skin inflammatory responses, and cardiac protection. It is highly expressed in the rods of the mouse retina, serving as the primary phosphorylation site for rod dendrites. In CD2/PKCα transgenic mice, overexpression of PKCα is linked to immune senescence.
