Target Information
Protease Activated Receptor 2 (PAR-2) is a receptor for trypsin and trypsin-like enzymes that is coupled to G proteins. Its function is mediated by activating several signaling pathways, such as phospholipase C (PLC), intracellular calcium, mitogen-activated protein kinase (MAPK), I-kappaB kinase/NF-kappaB, and Rho. PAR-2 can also be transactivated by cleaved F2r/Par1. It plays a role in modulating inflammatory responses, regulating innate and adaptive immunity, and acting as a sensor for proteolytic enzymes generated during infections. In mice, PAR-2 is deeply involved in inflammation, immune response, pain perception, and intestinal barrier function. It is expressed in the colonic epithelial cells of mice and regulates intestinal permeability through protease activation. Studies have shown that proteases, such as trypsin, activate PAR-2 by cleaving its exogenous amino terminus, exposing its binding site, and subsequently increasing intestinal permeability.
