Target Information
Proprotein convertase subtilisin/kexin type 9 (PCSK9) plays a vital role in maintaining plasma cholesterol balance. It interacts with members of the low-density lipid receptor family, including the low-density lipoprotein receptor (LDLR), very low-density lipoprotein receptor (VLDLR), apolipoprotein E receptor (LRP1/APOER), and apolipoprotein receptor 2 (LRP8/APOER2), facilitating their degradation within intracellular acidic compartments. As part of the protease family, PCSK9 decreases the number of LDLRs in liver cells by binding to them and targeting them for degradation in lysosomes, which results in elevated levels of LDL cholesterol (LDL-C) in the bloodstream. In mice, overexpressing either the wild-type or mutant forms of PCSK9 significantly reduces LDLR protein levels, leading to an increase in plasma LDL-C levels.
